SEQUENCE ANALYSIS OF CHLOROPLAST 16SrDNA-RPS12/7 AND ITS
SPACER OF DENDROBIUM ORCHIDS

      The 16S rDNA-rps12/7 and its spacer region of the chloroplast DNA were isolated from two orchids, Dendrobium Jaquelyne Thomas and Den. Ear Sakul. The amplified fragments of 3,782 bp from total orchid DNA extraction were sequenced and evaluated. We found that 16S rDNA-rps12/7 nucleotide sequences of two orchids were identical. Surprisingly, the alignment of their nucleotides with other published sequences show that orchid chloroplast 16S rDNA and its inter spacer between 16SrDNA and rps12/7 are similar to dicots, Amborella trichopoda and Nicotiana tabacum but differ from other monocots including Triticum aestivum, Oryza sativa and Zea mays. According to the high conserve sequence in 16S rDNA-rps12/7 region, it is possible to integrate this region into vector cassette of tobacco plastid transformation vector and apply as a vector for orchid transformation.
